The subfamily Letovirinae was established for Microhyla letovirus 1 (Figure 1 Letovirinae) based on DEmARC analysis of the nidovirus-wide conserved 3CLpro, NiRAN, RdRP, ZBH and HEL1 domains.

As only one genus is currently recognised, the genus description corresponds to the family description.
Viruses with genomes that differ in their nucleotide identity by more than 5% over the whole genome are assigned to different species.
Viruses assigned to the genus Amplylivirus form a distinct monophyletic group based on well-supported Maximum Likelihood or Maximum Clade Credibility trees inferred from complete L sequences. Viruses assigned to the genus have been detected in amphibians. Amplyliviruses most closely related to replyliviruses, which have been detected in reptiles, and lyssaviruses, which infect mammals.
